Modulation of Autophagy, a Therapeutic Approach to Mutated NRAS

Abstract

Here we discuss the mechanistic relationship of autophagy to apoptosis and specifically address the challenges in understanding the therapeutic
approach to RAS mutated cancers, through modification of autophagy. We then present a case report of NRAS mutated positive liquid biopsy (on a
patient with possible neuroendocrine tumor), and a treatment approach consisting of modulation of autophagy. We explain the mechanisms involved
with tumor mutated allele frequency response in liquid biopsy and conclude that such approach could present as a viable option in RAS mutated
tumor therapeutics and a basket trial recommended for future clinical validation.
